The instruction to "blow sample" helps to break up any cell clumps that may have formed. This action helps ensure that all cells are evenly dispersed in the electrolyte solution, leading to a more consistent sample and a more accurate count. Ultimately, the goal is to have a sample where each cell is in a single suspension, such that it allows for accurate detection and counting. 

The principle of the JimBio cell counter is electrical impedance. When the cell

passes between the two electrodes, it blocks the current momentarily because the intact cell membrane is an electrical insulator. This interruption causes a temporary increase in electrical resistance, or what is called impedance between the electrodes. The instrument detects the increased resistance, which corresponds to one cell, and counts these events. 

So, it is important that you have single cell suspension for accurate counting.
