I am performing siRNA transfection to inhibit a certain matrix RNA. Does it make sense in this case to determine the reduction of this RNA by reverse transcription on PCR?
You would need to use QPCR. Yes you do need to check how much knockdown you are getting with your siRNA.
it's possible to do an RTqPCR in order to see the KO of your RNA, using the adequate controle
Raoudha Missoum Thank you for your answer! Do you mean i have to use a scramble siRNA as a "-" controle?
I haven't used a scramble siRNA but i used 2 housekeeping genes at the same time (GAPDH & ActinB)
