Once the plant cells are broken open through liquid nitrogen treatment and grinding, various cellular proteases can release and degrade your enzymes. So, specific chemicals such as anti-protease agents need to be added. Check out some protein/enzyme purification articles to read and find out.

If "molecular assays" means analyses on DNA and RNA, be aware that grinding after freezing in liquid nitrogen is the most used and efficient method for nucleic acids extraction. If interested in RNA extraction, be sure to add proper buffer just after thawing or to the frozen powder. The same for DNA, but it is much less sensitive.
