First, please define a specific bacteriophage and its host bacteria. For example, coliphages which reside inside Escherichia coli. For the isolation of coliphages, the soil near hospital drains are preferred. You may take 1gm of soil sample, dry it and make serial dilutions at 1: 10, 1: 100 and 1: 1000 or more. Filter the the debris and soil. Take only supernatant. Then you prepare a general media like Nutrient agar and inoculate the supernatants onto the agar plates. You may use pour plate or swabing method. Then incubate the plates in an bacteriological incubator. After 12-14hrs of incubation, check for bacterial lawn along with clear zones (plaques) in the plates. The presumptive bacteriophages should be visualized using an electron microscope.
