I have been culturing iPSCs that i reprogrammed myself for several months now. I have had some contamination in the past but that's been fairly obvious. Cloudy overnight type of stuff. This is different and I am unsure if it is just strange debris or contamination.
I am referring to both the strange dots on the plate surface and the plastic looking structures. I suspect it might not be contamination but simply that my culture is not particularly healthy.
Additional helpful info:
1) cells are feeder free! cultured on matrigel and fed mTeSR
2) the dots on the cell plate have been there for a few days without change, the iPSC colonies look fine!

Hi, it looks like plastic structures from your pipette.. look at the media the iPSC is growing, if its cloudy then I am sure is contamination. The dots are death cells of iPSCs. Otherwise, your cells are growing happily :)
Hi there, we've been having the same issue and believe it's to do with the Matrigel. Going to try filtering the Matrigel-DMEM/F-12 mixture before coating plates to see if it will help, but if you found another solution, would love to hear it!
