Dear all, could you please highlighten me if there's cell culture contamination in my ARPE-19 cells? I noticed there's a bald spot in the T75 flask and it seemed the ARPE-19 were growing around the spot. The cells tend to grow at a slower pace too. Thank you.
- Seemed like you injured the flask surface by your pipette during splitting or perhaps the flask was not appropriately positioned in the incubator.
- This is sometimes observed while seeding/ splitting in a low volume of media.
- I would suggest to transfer some cells to a new flask and let them grow for a while. Nevertheless, check you incubator shelves, test regularly for mycoplasma contamination, and split your cells in at least 15ml total volume in the T75 flasks.
Given the circular shape of the low-density area, I suspect that was a dry spot when you initially set up your flask. The medium did not cover it until later, possibly after feeding or examining the flask. After that, a few cells seeded the area.
It is important to use sufficient volume to cover the entire floor of the flask, and I always spread the culture medium by gently rocking the flask until all the surface is covered.
Another possibility is that the flask was not uniformly treated during manufacture and the cells are only adhering to the treated areas.
