I want to do eplet matching using HLA matchmaker, however, I have only 2 digit HLA typing available with me dont have MFI values. How should I go ahead with eplet matching?
In my eyes it is not possible to corelate "2 Digits" (today: first Field) typing resolutionwith eplet matching. You need "4 Digits" typing resolution (today: typing of the first and second Field) at least.
I have tried using Haplostat software to convert 2 digit typing to 4 digit typing.Now is there a way to perform eplet matching with these 4 digit typing results obtained from Haplostat.
Since 2010 we have a new HLA nomenclature (WHO) in the whole world!
Terms like 2 and 4 digit resolution are obsolet!
I am not sure, if Haplostat will give you the right translation from the "2 Digit resolution" results to the most presumptive "4 Digit resolution" result in your population.
To give an exsanple: B*15 in Caucasians, Haplostat would this translate into B*15:01. But in Asia (India) the translation should be B*15:02.
If you know your population and most frequent alleles then you can estimate the 4-digit typing and use this data to look at eplet mismatches. Its not 100% but we have done the analysis in Australia and show good correlation with actual 4-digit typing. Kind regards.
