I have used the gelatin coated slides for mounting the polymer containing cells on it (which was post fixed with paraformaldehyde). But while staining, it gets peeled off. The polymer is resistant to alcohol but the sections get peeled off in hydration and dehydration steps. Any tips?
This may be a significant problem which depents on the tissue type, the staining protocoll and the glass slides you use.
To get good results without peeling off you may use glass slides "Super Frost" or "Super Frost Plus". If you stain a very dense tissue a thickness of max. 3-5 µm of the slides would be good. Last but not least you may try to dry your slides after fixation.
Is it will be good for even synthetic polymer based cell culture sections
I have seen people using gelatin as well as superfrost slides. Is the adherence will be be better in super frost plus slides
We use positively charged slide from Mercedes Medical and run the antibody staining while the slides are horizontal rather than vertical.
