For the detection of Th17 cells, I am using IL-17A antibodies from BD Biosciences in the patients with Hyper IgE syndrome.But my results (percentage of IL-17 cells) drastically vary from sample to sample.
I want to know is there any other marker or antibodies for detection of IL-17A, apart from IL-17A from BD biosciences by using flow cytometry
First I gate for all CD8-CD4+ cells. Next, I take all CD45RA+. Then gate for the CCR6+ cells and finally the Th17 are CCR4+ CXCR3-. Your Th1 and Th2 will be comprising the CCR6- population (Th1=CXCR3+CCR4-; Th2=CXCR3-CCR4+).

Are you referring to different clones, or to another way of detecting IL-17? Regarding IL17-A, there are many different clones such as N49-653 and SCPL1362 (both BD), or eBio64DEC17 and 20LJS09 (Thermo Fisher), all of which work well.
Alternatively, you can detect IL17-F (clone O33-782), which is often, but not always co-expressed with IL17-A.
But for human samples it is not unusual to observe large sample-to-sample variability.
