we use the following procedure   for the extraction of both  ABA  and cytokinins: extract  fresh plant material or freeze dried material into 80% methanol for 48 hours at 4C in darkness. then combine the  extract and evaporate to aqueous phase using rotatory evaporator  at 40C . then ,make  to 50 ml using distilled water. make the p H of extract  2.5 with dilute HCl. Do partitioning with ether for 3 times. combine the  ether phase  extract and evaporate to 5ml using rotary evaporator. this fraction contains ABA. then,  make the aqueous phase pH 8.5 using dilute NaOH. do  portioning with ether, three times , combine and evaporate to 5ml. this fraction contains cytokinins. then , do quantitative determination of both ABA and cytokinins   by higher pressure liquid chromatography (HPLC). 

ABA you can extract in 80% methanol and there are many methods for the extraction and purification of cytokinins are available in literature.

ABA abd auxins can be co extracted and purified
