We have to identify candida surface antigen to which our novel recombinant candida antibody will bind. I have gone through various articles but unable to find simple in vitro assay protocol for antigen identification. we did western blot using candida lysate and our antibody.The band of same size, identified in WB, were cut from SDS-PAGE (running parallel) and did mass spec. But we get lots of hits in mass spec. Now our further step is to identify the antigen to which our antibody will bind. Is there any in vitro assay for it? Please suggest. Thanks. Ajit
The usual method is the pull-down assay, in which the antibody is used to immunoprecipitate the antigen from the lysate. A control sample is prepared from which the antibody is omitted. The proteins pulled down by the antibody are then identified by MS.
