I used urea complexation procedure to concentrate polyunsaturated fatty acids (Omega-3 fatty acids) of fish oil following the attached protocol. Starting oil contained around 10% omega-3 fatty acids which was increased to 65% after urea complexation. There was no significant oxidation problem as fatty acids in organic solvent did not rancid very fast. I suggest you to follow this protocol.
The clathrate formation of urea with saturated unbranch fatty acids it is an excellent method for purifying polyunsaturated fatty acids. The method suggested by Monjurul it is ok. If your polyunsaturated FA it is very sensitive use absolute ethanol, try to do all the procedure in ambar material to avoid light and in glassware with taps.
