I would like to know, if I do a co-culture of lactic bacteria with Bifido, can they be differentiated on the basis of any bio-chemical test?
as PCR can easily differentiate them, but any other easiest method?

For enumeration, you can try to do an antibiogram for each bacteria and then add the convenient antibiotic at the convenient concentration in the solid medium.

2. another method is detection of fructose -6 phosphate phosphoketolase assay - specific for bifidobacterium

You can design fluorescent DNA probes or peptide nucleic acid probes targeting 16S rDNA sequence. Hybridize these probes with bacteria. This will help to study and differentiate co-cultured bacteria.

have you tired TOS-MUP agar for bifidobacteria (ISO 29981:2010) and MRS/CL/CIP agar fort L acidophilus(ISO 20128:2006)?
