Suppose the column packing flow rate is 30 cm/hr and the recommended sample run flow rate is 15 cm/hr; the resin pressure is maximum 0.02 Mpa. As the size exclusion chromatography requires longer time so is it a good decision to equilibrate column with 20-25 cm/hr. Thank you.
Probably not. Most problems with chromatography occur because the column has not been properly equilibrated so the retention time will shift. I would follow the column's manufacturer's directions.

Well, that sentence does not make sense as written, but YES, the packing material can be damaged by running too high a flow rate through it. Column equilibration is a process (process of stabilizing and reaching an equilibrium), not a flow rate.
Be patient. Sometimes these things take time. Equilibration (by definition) means you run at the SAME flow rate as your analysis method, not a different one. Running at a higher or lower flow rate changes the properties and access of the fluid to the support, so results in different conditions.
Never run at or near the original packing flow rate too (as Mangal mentioned). Changes or damage to the support and/or packing conditions can result (BAD).
Thank you BIll for your valuable suggestion regarding my question and sorry for the inconvenience. In case of size exclusion chromatography the recommended flow rate for sample run is half of the packing flow rate as per the resin manufacturing guideline. Sometimes if the column stored with 20% ethanol then for washing and equilibration it takes 3CV, and for long height column I experienced the equilibration time nearly at 20 hours.
After searching and watching some webinars I got some suggestions that I can equilibrate the column to increase some flow rate by carefully look after the resin pressure.
Here, Mangal Singh said we should not exceed 75% of the packing flow rate though back pressure is crucial. But, if the back pressure is not created then will it be a good decision to use this flow rate?
20% EtOH has higher viscosity than water so you should be even more careful with the flow rate. As others have stated it is not the flow rate per se that will affect the packing but it is the dragging force excerted on the beads. For the same flow rate that will be different for different liquids (and temperatures). The time you might save will not make up for the time you have to spend repacking. You should just accept that size exclusion is a time consuming technique. Fortunately you do not have to sit and watch it.....
