It well known that, the protocol of Triton WR 1339 induced hyperlipidemic model is the standard method to screen the herbals for hypolipidemic action. I have read several papers in which there are no clear-cut instructions. For example, this is the succinct protocol:
I) It has been instructed that, animals must be kept in fasting for at least 12-15 hours.
II) On day 1, a Triton injection (i.p) was given and after that, the selected drug will be given.
III) After 24 hrs of Triton injections (Day 2), blood sample 1 will be collected. After the 1st blood collection, another dose of our target drug will be administrated.
IV) At the end of the 48th (Day 3) hour, the 2nd blood sample will be collected.
From the aforementioned protocol, I understand that animals must be kept fasting (waster is allowed) nearly for 63 hours. Can animals survive with this prolonged fasting? Are there any articles, which has a detailed protocol for this method?.If yes, please share the link that demonstrates this.

The peak serum lipid level is attained in 18 to 24 hours after which the serum level slowly returns to normal from 36 to 48 hours after Trtion WR-1339 i.p. injection.
Hence, the test can be stopped at the peak interval once validated with all the time intervals. The two intervals with 18 and 24 hours can be proivded where the effect of test substance can be compared with non-treated group. As the peak level of serum lipids by itself goes to decline phase after 24 hours, the target test substance effect cannot be compared directly with non-treated group. Hence, single dose study would suffix for screening hypolipidemic models using Triton WR-1339 induced hyperlipidemia.
Also, animals can be fasted only for treatment (initial 12-16 hours); during the study feed can be provided, as it would be common for all groups. Normal rodent feed in India usually contains 4 to 5 % kcal fat.
