It depends on the receptor. Pertussis toxin can be used in vivo and will ucouple Gi/Go linked GPCR. However, Ptox has plenty of additional off target effects. I don't know of any other agents that will do what you want. I would imagine that any agent that does this will have dozens of off target effects that may complicate any interpretation of your observations.
