Since different groups reported different markers for the same type of cancer, you better use a panel of markers especially picking up those markers who have been reported by more than a single lab.
How about "Hoechst 33342 Staining for Side Populations"?
Did anybody use this method as a way to assess the alteration of "the stem cell portion" in whole cancer cell population after certain treatment?
There have been many who used the SP/Hoechst method, but the functional assays remain the eventual confirmatory step by showing the ability of the SP for achieving serial passages of tumors in mice, the differentiation potential and the sphere forming assays. Some looked at the molecular targets of chemotherapy in the SP cells (e.g. http://mct.aacrjournals.org/content/10/2/325.long)
The "gold standard" is in vivo tumorigenicity in immunocompromised mice. Chemoresistance is another functional characterization of CSCs.
