And what does it mean when the first two peaks of my titration are endothermic? I'm titrating a substrate to a protein and it was supposed to bind but not react. The MicroCal PEAQ analysis tell me it's not binding, though.
Also, the baseline cannot be adjusted at that point and I think this might be prejudicing my analysis.

Apparently you are having technical problems but, on the other hand, what do you mean with substrate if it does not react? A substrate by definition reacts. I would think that the protein is in fact an enzyme. Also titration means that you expect more than 1 substrate molecule binding to one protein molecule? Do you have the data from a "control" with another substrate and the same protein?
