I need to acquire a human cell line that both genetically and physiologically represents VAT, and differs from other tissues, such as subcutaneous adipose tissue and brown adipose tissue. Also, it cant be a primary culture, as they have shorter life spans
The best established cell lines is the 3T3-L1. It has many short-comings but it is a good starting point. An alternative is the AP-18, but has not been used as extensively.

Yes, I studied the SGBS cell line, but it has been recently described that it presents a heterogeneous phenotype, which transitionally changes between brown and white adipose tissue, and I need a cell line that would specifically represent visceral adipose tissue, and phenotypically differs from subcutaneous adipose tissue (both are white adipose tissue subclasses)
You will find that this is a problem with most culture cell lines.
One person that may know if there is such a cell line would be Nick Hastie from the
MRC Human Genetics Unit, Institute of Genetics and Molecular Medicine (University of Edinburgh). They are doing some very interesting work on visceral vs subcuteneous AT; published a very interesting paper entitled "Visceral and subcutaneous fat have different origins and evidence supports a mesothelial source"
Dear Luis, I would recommend the SGBS cells from Martin Wabitsch's lab as model for subcutaneous AT (SAT). We have made several publications with these cells, specifically regarding adipokine research. The adipokine profile overlaps for >80% with the adipokine profile of SAT tissue biopsies. However, a similar type of cells, closely resembling visceral adipose tissue (VAT), is something we are looking for too. VAT preadipocytes are commercially available, although the costs are quite high and propagation of the preadipocytes is possible for only 2-3 generations as you already indicated. Not really a 'cell line' type like SGBS. So, if you may find VAT 'cell line' cells I would be happy to hear it.

yes, I also recommend SGBS cells. But there is a possibility to work with primary preadipocytes (PMID:18198300). I worked in this lab and I used the PM-4 medium to passagge primary preadipocytes for several times. They will still differentiate. However, it depends on the donor and adipose tissue site how well they will differentiate. But it is definitely an option!

SGBS cells have been isolated from the subcutaneous adipose tissue of an infant with Simpson-Golabi-Behmel syndrome (SGBS). They are not immortalized or transformed and retaint their ability to differentiate into adipocytes for more than 50 generations.
As thy are dependent on a PPARg agonist during adipogenic differentiation, they express UCP1 to a certain amount.
To my knowledge, there is no visceral preadipocyte cell line available - there you have to stick to primary cells.

There are 2 known human adipocyte cell lines, Lisa-2 from liposarcoma and the SGBS which we have used from SC fat of WAT from infant. SGBS has a good adipose differentiation even upto 50th passage. I believe there was also an immortalized primary adipocyte but only available to order from Japan (I believe that is SC too not visceral).

Hi Chenji Ye, Yes i have used before this cell line and also still continue to work with this cell line. We used all mediums and reagents from compnay that reccomended because this cell line a little bit to hard for working like attachment to flask and proliferation were problem.
