Is there a recommended assay for ralstonia solanacearum growth curve in peppers?

I want to do a growth curve of Ralstonia solanacearum in peppers. There are several protocols that use stem sections or leaf discs for sampling. Which would be a preferred assay for peppers? Also, what O.D of bacterial suspension is recommended for Ralstonia growth curve analysis?
