Usually after cryopreservation the cells will have reduced viability. But for the first time, I found an increase cell number. I used 10% FBS, DMEM and DMSO for freezing. There was a 17% increase in the cell number.
It is not possible to get increased cell count after cryopreservation as the cell freezing media and the ultra low temperature does not support cell division. The possible cause for getting higher cell count after cell freezing is probably erroneous counting at the very first instance when the cells were frozen. If the cells after trypsinization do not detach properly and if there is incomplete trypsinization the cells may not be available as single cells. The possibility is that what was counted before as single cells were actually a cluster of cells and after freezing those cells became dissociated from each other and became appear as single cells when recounted after freezing.
