I'm trying to put a split GFP onto the very N-terminus of the BKa channel (gene slo-1) in C. elegans, which is a membrane protein. There is no signal peptide but rather a property of one of the transmembrane domains ensures that it goes to plasma membrane. Problem is if I put a 700 nt split GFP onto N-terminus, the N-terminus probably won't go inside the ER lumen and thus probably won't be extracellular anymore. I need the split GFP on the extracellular side on the end of the N-terminus. Thus, is there a sequence I can also add to ensure that this split GFP N-terminus is inside ER lumen and thus is later extracellular? I was thinking about MyrPalm split GFP but I don't think MyrPalm works in C. elegans. Or does it?
