I am looking for a good way to concentrate a large volume of allantoic fluid to a smaller volume to increase the viral titer.
Are you concentrating >1 liter? If so, a simple method of concentration would be by ultracentrifugation:
1. Clarify allantoic fluid by low-speed centrifugation at 1200 x g for 10min at 4-10°C (can use refrigerated table-top centrifuge)
2. Centrifuge clarified allantoic fluid in ultracentrifuge at ~45,000 x g for 3 hours at 4-10°C.
3. Decant (remove) supernatant.
4. resuspend pellet in Calcium borate buffer (to retain NA activity).
5. PBS may also work; however, we only perform this when we are concentrating virus for vaccination and are not worried about live virus.

If you are getting low influenza titers from eggs, you can try different MOIs. Our lab has shown lower MOIs actually increase viral production in eggs.
