you have to coat the surface, eg. with collagen - I've done this before and it is ok. There are some commercially available kits for such coating.
Do yoou have a protocol that you can share with me. I also appreciate if you give some commercial kit names.
I used coating medium from life technologies (matrix coating kit cat. no. R-011-K), but I think there are many available in other vendors. There are also specialized media for keratinocytes, also with culturing protocols included.
Best stratification of primary corneal epithelial cell cultures is achieved when growing on a permeable membrane in the presence of an underlying layer of fibroblast feeder cells. Generally murine 3t3 cells are best. If seed early passage cultures around 50-100 cells/sqcm can generally achieve good results within 2 weeks.
