I want to cultivate a fungus from a wood sample I collected with an increment borer. This is an ascomycete that resides in the tree core until the temperature rises and then it travels to the tree bark where it starts producing spores. I want to isolate the fungus so I can see if a tree is infected before it starts producing its spores.
When placing these samples on a PDA petri dish I found that it becomes overgrown with different fungi. Is there a method I can use to prevent the growth of other fungi, or a different method I can use?
Renee Klaver, after following all Kouadri Mohamed El Amine said you have to first culture on water agar first before inoculating onto PDA. This helps to eliminate saprophytes that outgrow the actual pathogen. This approach will be more appropriate than inoculating onto PDA directly.
You have to do series of isolation to establish the actual organism and you have to also conduct a pathogenicity test

Then You can also modify the PDA medium, by replacing Dextrose with Sucrose.

The moist chamber is a very suitable method to isolate the fungi associated with wood. So, when the fungal growth appears, it can be transmitted to a suitable medium.
The sample must surface sterile with NaOCl 10% from the trade solution for 2-3 minutes, then it must be washed with sterile distilled water to remove the effect of sterilization. After that it must dried with filter paper, cut to small fragments and cultured on PDA or any other medium. Finally, the plates incubated under 25 C for 3-7 days.
Yes, the modification of PDA medium by replacing dextrose with sucrose is benefit. But you must use half amount when you use sucrose. You must add 10 gm sucrose to the liter medium instead of 20 gm of dextrose.
