Hello everyone,
I would like to extract tRNA using TRIzol (from ambion) and to get the methionine-tRNA (with the amino acid attached). I know that tRNA is extracted using the TRIzol reagent but am not sure whether the ester bond between the tRNA and methionine can sustain the extraction (especially considering that the methionine-tRNA linkage is the weakest of all amino acids).
Does anyone has a working protocol for this extraction?
EDIT: I don't need a pure tRNA-Met, I just need a good portion of it in a total RNA extraction. I plan on digesting the RNA with RNase. The end goal is to know that the methionine in the solution after RNase treatment came from tRNA and not other sources.
