Ekemini Obok Yashwant Chavan I have used the miniprep from Qiagen a lot, but it extracts the total DNA (i.e. both fungal and plant DNA at the same time).
Hello, by benzylchloride method you can extract both plant and fungi DNA by same procedure and same reaction mixture and you will receive both DNAs mixture. If you want to isolate only fungi DNA the better way is to isolate fungi culture from your material as pure culture and then perform DNA isolation.
I have done separate isolation, but a friend mentions that there is a possibility of isolation only plant DNA; I do not think it is possible; that is why I am asking.
I think that whenever you do the extraction of infected plant material you will get DNA from the plant and the fungus. If you want only the DNA of the fungus try to make isolation. If your goal is to detect the fungus in the infected material, check if there are specific primers for this fungus and you can use the DNA of the infected plant material. In this case and if the specificity of the primers has been tested against the DNA of the plant and does not react, you will only get a band if the fungus is present.
As far as I know, I think it's not possible. If you want only the DNA of the fungus, try to isolate the fungus in culture medium and then do the extraction of the culture
