We are performing enzymatic reaction adding the cofactor for that enzyme. Without adding the cofactor the assay condition is suitable.
Can you give some specifics about the enzyme and the cofactor?
The name of the enzyme is cysteine synthase and the cofactor is PLP.
In that case, the cofactor (or coenzyme) is essential to the catalytic mechanism of the enzyme. No reaction can occur without it.
