There are several nitric oxide kits produced from different chemical companies, that is tempting to save time and looks promising as a valid method to determine nitric oxide. I would like also to exchange our knowledge about using nitric oxide kits for determining nitrate and nitrite in food samples and biological fluids. Furthermore, any one has experience using urinary dipstick to determine nitrite in urine samples.

This nitric oxide assay kit is for quantitative determination of nitrite (NO2-) and nitrate (NO3-) in biological fluids. The kit uses the enzyme nitrate reductase to convert nitrate to nitrite. Nitrite is then detected as a colored azo dye product of the Griess reaction that absorbs visible light at 540nm. The interaction of nitric oxide (NO) in a system is measured by the determination of both nitrate and nitrite concentrations in the sample. The relative levels of nitrite and nitrate can vary substantially, therefore the most accurate determination of total NO production requires measurement of both nitrate and nitrite.

Urinary NOx has been generally accepted as a measure of total body NO production. The proportion of NOx excreted in the urine constitutes 50 to 60% of the total body clearance of these compounds, with the remaining amounts being eliminated in unknown proportions by the exocrine glands and by the respiratory and gastrointestinal tracts.
The total amount of NOx measured in the urine and serum derives from both endogenous and exogenous sources. Some studies demonstrate a significant decrease in NOx in urine and serum after 24 hours of a NOx restricted diet. You have two ways either do it by kits or prepare your chemical staff by your self. I have attached a classic method to determine NOx in several biological fluids.

Dear Manal,
I have noticed that you have also asked for dipstick for urinary analysis. I would not recommend to use these strips for NOx it is indicate whether a positive or a negative result (pinkish red colour for nitrite) that gave you overall idea. We usually use this in our hospitals as a quick method for nitrite and other parameters with no exact values.
