I isolated some PBMC and they are in liquid nitrogen now. I've found online that people could differentiate monocyte from PBMC to macrophages, which is the MDMs, but I was wondering if I could have the alveolar macrophages from PBMC as well? Thanks!
Alveolar macrophages (CD14 low, CD16 high, CD71 positive, high MHC class II ) are short-lived cell, continuously replenished by the circulating pool of monocytes. I believe it is possible to generate Alveolar macrophages (AM) from peripheral blood monocyte derived macrophages (MDM) treated with Granulocyte-macrophage colony-stimulating factor (GMCSF) which plays important role for the differentiation of AM.
Yes, circulating monocytes can travel into the lung tissue and populate the alveolar space (mostly during some sort of pathological event). But TRUE alveolar macrophages do not originate from the bone marrow. While there might be still some debates... I'm pretty sure most have come around to the idea of the fetal origin for tissue resident macrophages.
True alveolar macrophages are actually quite long living. They are also express high CD11c, and probably also CD163.
