Dear researchers,
I understand that we can get drugs from polar compounds, but these are usually hardly bioavailable. This means the drug has to keep being modified to improve it's bioavailability (structurally).
Is it worth looking into for a PhD considering these challenges?

Consider the bunch of research devoted NOT to find new API, but to find the way to get them more bioavailable, more stable... most of pharma patents now are on these arguments. And there is a lot of multidisciplinarity, since biochemistry, pharmacocynetic, molecular chemistry and much other are involved - there is enough for many PhD, regardless the starting point whether a physician, a biologist, a chemist or a pharma tech.
Are you a chemist? Generally, it's predetermined by what the target allows, and how that aligns with ADME SPR, but there may be a strategy in prodrugs to increase permeability. If so, this should be adopted up front. Abbvie is currently touting their success with developing a Bcl protein-protein inhibitor although it was labor and resource intensive so just a belief in tenacity isn't enough. It's all about dose, potency, and tolerability.
The field is moving. You will be surprised what a hungry PhD student can do.
Yes, it is worthwhile. Polar compounds can efficiently reach tissues by loading them on different carriers. It is no more restricted to only structural modifications.
Thank you colleagues for your kind responses. They are an eye opener to me. I used to avoid polar compounds in drug designing as my prodrugs, afraid of a lengthy expensive process of modification to improve two things: Bioavailability and stability, but now, you have given me a different view

Yes I sure am a Chemist, PhD researcher in designing drugs. Now I have a better feeling about polar compounds as potentials that can lead to stable and bioavailable drugs. I have not yet explored carriers, I am looking at them now.
