We have a sonication bath, but I was just wondering if purchasing a probe sonicator would lead to better disaggregation of viral particles for both purification and electron microscopy purposes, or will there be no difference at the same frequency? My main concern ss the container reducing efficiency, or is this not the case?
If you are only trying to seperate aggregates in a virus containing solution, then a sonication bath can suffice (depending on the virus, the origin of the solution etc.). If you are trying to purify virus from cells or organs then you need a sonicator with special probe for isolated tubes. We use a VibraCell sonicator with sucn probe from "Sonics".
