Is it possible to use PPM (TM) with antibiotics to eliminate endogenous bacterial contamination ?
PPM Alone or even Antibiotics alone can't really give a good percentage of endogenous contamination free explants for mass propagation . i am interested to know if PPM and antibiotics can be used in combination to increase efficacy of this treatment.
Dear Mohammad Sadegh Bagheri Ardabilian it's a good question. For sure you'll have a lot of answer to your request. The literature is plenty of references regarding the use of PPM for sanitizing. In my experience it doesn't work, even using greater doses than recommended. Since I don't observed any damage with these doses, it may suggests that probably can be used PPM combined with antibiotics. My question is, what is the purpose to mix PPM with antibiotics? As many researchers know antibiotics are not very well effective to produce 100% bacteria free plantlets and normally a mix of different methods are used to produce clean plant materials from any bacterias . I will use mix antibiotics solution dissolved in DMSO to have maximum penetration of antibiotics to plant tissue and use PPM or similar preservatives in solution to suppress bacterias that are sink in liquid medium . May i can reduce ph of medium to use maximize pressure on bacterias .
Sometimes the problem is not the substance, but the method that is used for treatment. I'm not expert in endophitics; however, I'm dealing with it every day, as every tissue culturist. I see quite difficult to obtain 100% bacteria free plants. If your objetive is to micropropagate, my advise is to focus your attention in to control the contamination, not to produce 100% bacteria (there're also fungi....) free plants. Microorganisms have evolveld together with plants, so both need each other in a symbiotic relationship; therefore don't underestimate it.

I too cant get rid of a pink bacteria which might be inside Papaya. As you suggest: maybe some plants are used to a bacteria. Only we want it clean!

Several ways are used for clean endogenous bacteria, it is a big matter, but looking a kind of bacteria you may use special antibiotic mixed with PPM.

Dear S. Wenkart I don't have ever worked with papaya; however, a kind of pink bacterium is frequent in tissue culture. Its arise, at least in my experience, is associated with errors esterilization of culture media and with manipulation of explants. Can you share some picture?
Dear Perea Dallos Margarita I'm agree with you. Contamination is a big problem, and its sulution is also a big matter, since many aspects determine the final results. It's highly difficult to succeed considering only single factors. As many experts have recommended, I only have obtained clean cultures combining selection of the right explants, the use of appropiated antibiotics (in my case always Cefotaxime and Carbeniciline), the applying of chronic treatment, the continuos screening and isolation of clean lines.
In my experiences i use a mix of three Antibiotic (Rifampicin , Cefotaxime and Vancomycin) to cover all in one solution as pretreatment after sterilizing for 1 hour .I get good results for several month from this treatment but now i have infections again . I am working on more rare plants from Sansevieria Genus , they are rare and i have not a lot of explants . Sometimes we are in hurry to produce clean explants because controlling endogenous bacteria is not effective always .
Pink Bacteria that S. Wenkart have is a common infection and mostly endogenous but i managed to eliminate it with my antibiotic mix .
Dear Mohammad, the re-emergency is permanent challenge that those that work with tissue culture must face. Sometimes, hurry might drive to lose a lot of time and explants. The solution is not always the use antibiotics but to check to all the procedure to obtain clean explants.
