I want to record FT-IR of surfactant solution. I am wondering whether there will be a layer created at the interface of the liquid and the wall of the cuvette which might interfere the spectra. If I freeze dry the solution and the take the spectra of the powder, I am afraid whether the micelle will be disturbed during lyophilization. Please suggest an easy way to overcome this. Thank you in advance.
Dear Ranjit ;I don't know the nature of your solution which want to measure? but you can measure your solution after you measure the cuvette ( as background ) then you can measure your sample within cuvette . or you can pick up one drop of solution directly applied in ATR mode of FT-IR.
Quartz cuvettes usually not suitable for FTIR measurement. If you have aqueous solution, the strong absorption of water at the IR takes some more difficulty during the measurement. Maybe you can try ATR unit and using water background but the water peaks change when you measure it in solution. Because of the different OH bonds in a water-organic system you will have peaks where water has peaks after background removal too.
There are some paper about FTIR measurement of protein in aqueous solution. Search for it. It may help you.
