Has anyone experience with collection of human bile during bile duct catherization (ERCP) and extracting cholesterol and bilirubinate microrystals from it?
I would like investigate small-sized microcrystals and they possible interaction with cells.
Unfortunately, from what I have read the problem is nucleation of small particles so they form bigger particles.
Any experience on that one?
Yes. I used to obtain bile for analysis with a weighted duodenal tube. Of course, endoscope is more expansive and less confortable but can also suck of bile flowing out (with a very soft depression to avoid wall succion ). It is possible to contract gallbladder with sorbitol infused in duodenum.

The biliary microlithiasis may be involved in a variety of diseases of both the biliary tree and the pancreas including the acute recurrent pancreatitis, AP in the pregnants, congenital anomalies in children, and so on (see: PMID: 20339782; PMID: 20069159, etc.).
The succion should be as low as possible; ideally, if the bile flows itself.
As for the nucleation of bile microcrystals (if does any matter for a mucosal lining damage), I have not met any publication like that. That is most narrow and specific matter depending virtually on the sort of crystals (cholesterol, Ca bilirubinate, and others); on the succion pressure, the tube diameter, etc.
The only way that will solve the question is the de novo experimental work with tubes of varies width, material; different succion regimen; and variances in bile chemical composition.
The fact is that the nucleation of lypophylic substances in water interface is both natural and envitable a process. Whether the degree of nucleation depend significally on the relatively low endoscopic succion pressure, that is the interesting question you made.
