Is it possible to enrich/selectively isolate poly-dT+ cDNA from a sample of cDNA which was reverse transcribed from total RNA using a mix of oligo-dT and random primers?
Thank you very much for the answer, I was hoping it would be as simple as that. We are planning to do RNA seq, but did not perform polyA enrichment or rRNA depletion before reverse transcription, so I would like to perform an enrichment of cDNA derived from polyA RNA at this point. Poly-dT beads are easy to find, but I would need poly-dA beads, right? Any advise on where to find these?
I used Biorad's iScript cDNA Synthesis Kit (http://www.bio-rad.com/en-be/product/iscript-cdna-synthesis-kit?tab=Description), which uses an RNase H+ MMLV reverse transcriptase for first strand cDNA generation, so I'm guessing I am dealing with single stranded cDNA? Or do I see this wrong? :)
