HI. I want to do Oil Red O staining on mice liver tissue. I have either parafin embedded tissue or frozen tissues that i have frozen in liquid nitrigen after cutting from mice and then at -80. Can any one suggest me the better way to use these tissues for Oil Red O staining ?
You will have to use the frozen tissue. The process of paraffin embedding dissolves and washes away lipid, so there is little left in paraffin sections.
Liquid nitrogen frozen tissue should be O.K. for subsequent Oil Red O staining. You should try to use fresh cut frozen section. If you use old frozen sections they are probably dried out and only give a bad quality.
