i am trying to detect bacilysin by MALDI-TOF/MS in the cultural filtrates of bacillus strains but i am unable to detect it. is there any reason for not detecting bacilysin in crude cultural filtrates of bacillus.
Hello, Bacilysin is non-ribosomally synthesised dipeptide that consists of L-alanyl and anticapsin units linked by a peptide bond.
May be the component of bacilysin missing in the MS analysis was alanine. Analysis involving bacilysin, the lowest m/z by the mass spectrometer may be not low enough to detect alanine. If you have base peak m/z 182.1 instead of m/z 271.1 (the bacilysin peak), would represent a loss of one alanine (89 Da).
