THP-1 cells do not seem to grow well in DMEM. Within 24 hours, cells form clumps and most of them undergo cell death. If you are planning to set up an experiment using DMEM, try culturing THP-1 cells in high glucose DMEM and expose these cells for a shorter duration (less than 24 hours).

Thank you for the helpful suggestion. It seems that U937 have a more M1-like state. The THP-1 are quite slow growing comparing to RAW264.7, so I'll give it a try by going through series of HG DMEM exposure.
