Hi Vivek, what kind of bone-marrow cells would like to culture? The bone-marrow contains quite a variety of cells, including almost the entire hematopoietic lineage (i.e. lymphoid, myeloid and erythroid) and quite a few cell-types from the mesenchymal lineage (i.e. mesenchymal stem cells, osteoblasts and adipocytes). You can culture most of them to some extent, but all require different conditions. .
Thanks Hendrik, I am not trying to differentiate the cells into any lineage. I want to take the cells and maintain in the media. My question is Which media is better to maintain the cells isolated?
I isolate/expand stem cells from rat bone marrow with DMEM/F12 + 20% FBS + antibiotic/antimycotic. Freeze them down in 80% FBS, 10% DMEM/F12 and 10% DMSO. When you take them out of the deep freeze you can grow them in DMEM/F12 + 10% FBS + antibiotic/antimycotic. The stem cells won't differentiate.
