I am working with establishment of 3D culture of mammalian cells on porous membrane (0.4 micron pore size) using Matrigel. I am using thick gel protocol for the same. Considering the conditions I am providing, I need to wet the porous membrane prior to addition of Matrigel (containing cell suspension).
Will the Matrigel still form a thick gel on incubation at 37 degree Celsius after addition to the membrane which has been wetted with cell culture medium?

I have been using 5% Matrigel, prepared in the culture medium and coated on the cell culture plate for 1h at 37 degree Celsius. I believe the membrane lightly wetted with medium, taking care that it does not dilute the Matrigel containing cell suspension significantly, should not be a problem for setting the Matrigel at 37 degree Celsius.
