I am planning on getting histology sections from a very small-sized sample.

The section must show the center of the tissue, so I have to divide it into halves as accurately as possible before I immerse it in formalin.

Because of its small size and squishy character, it is very difficult to achieve a perfect half.

So I figured, if I snap-freeze it with liquid nitrogen, it would be much easier to break it into even halves.

But my worries are that there might be some possibility that the freezing process, although short, might disrupt the tissue structure very seriously.

It would be better if i just fix the whole tissue in formalin and just section it out until the apex, but I have to save the other half for other analyses.

Does anyone have any experiences regarding handling small tissues?

thank you.
