I have extracted genomic DNA by conventional CTAB method. I need to do partial sequencing , for which, I was asked to purify my DNA using spin column. I have checked the purity of my DNA using nanodrop and got the ratio as 1.78. Am I supposed to purify my DNA with spin column or is this sufficient enough.
What is the concentration of your DNA, Spin columns does well purify the DNA but loss of DNA also too high, therefore depending upon your initial DNA concentration and purity do prefer column based purification.
Sequencing always works better if you purify your DNA/ follow the instructions from company (about concentration etc.). Maybe run DNA also on an agarose gel to check the quality, i prefer it instead of NanoDrop because you can better see contaminations with RNA/proteins.
