Yes better should not, when you pour media you have to careful cool media in plates and after dry keep plates opposite side with nice rapping. Shilpa Devkota
To avoid the drops formation, you must cool the medium to a suitable degree before pouring, then you can pour the media carefully and left the plates opened for about three minutes in the sterilized hood, and you must incubate the plates with reverse side.
Thank you everyone for your kind suggestion. The lab that we are working is not that much well facilitated in order to keep it perfectly aseptic. Can we pour the PAD on plates and then left to cool the media for about 3 min with opening the UV light in laminar airflow?
Thank you sir, will it perfectly work to keep the plate free from contamination while keeping it with 20-30 % lid open to cool down the PDA. I am scared, whether contamination could occur while opening the lid.
I think is not mandatory. But if you let water drops in the cover of your plate this can lead to contamination because the water can carry other organisms.
I have this problem all the time.
You can always let the plate lid open in the laminar flow hood to avoid the drops and after that put in the U.V. light for 15 min.
If you problem is after your inoculation you can put the lid next to a flame very quickly to evaporation.
