I'd like currently to evaluate virus-induced NLPR3 inflammasome activation both in vitro and in vivo. For in vitro analysis by Western Blot, there is no problem. However, I did not find important information about identification of NLRP3 activation by immunohistochemical staining in mouse tissue sections. Is there somebody having experiences on this method? Thanks a lot!

I think u can detect both proIL1B and secretory IL1B.
As Inflammasome mediate the activation of caspase-1 and the cleave pro-IL-1β and pro-IL-18 into their active, secreted form. So if inflammasome is active u can detect more phosphorylated caspase1 and further secreted IL1B in supernatant. For tissue samples you can also go for Immunohistochemistry.
