1. Why you not to normalize the sample data?

2. If you normalize the sample data, how can you perform the statistical test, i.e. what is your preassigned value for comparison. It should not be zero and it should also normalized, how? Otherwise, the sample observations and the preassigned value are not comfortable.

Sorry the question might not have been very clear.

Normalization was done for a PCR data, which is quite normal.

Anyway, I did a little bit of digging about this and somehow got an answer.
