that depends. We used a 32-Multiplex from Porcarta to analyze cytokine secretion from macrophages in vitro and we decided to make technical duplicates already in the experiment itself and accordingly we transfered this duplicates to the Procartaplex.
However, for in vivo samples, like tissue lysates, blood plasma and so on, we made monoplicates, with one sample (representing one mouse) in one well. Since a sample from one mouse (or human in your case) represents one n, there is no nesessity for technical duplicates of the same sample.
