I have to order dsRNA (25nt) and I asked for a quote. The supplier insert HPLC purification that is very expensive. I would like to know if it's strictly necessary. No HPLC purification would affect negatively my experiment?
if you just want to produce siRNA- no need for HPLC purification, just mix strands in 1:1 ratio and thats it. small excess of one strand wont make a difference. for in vivo studies of course you want to ensure your siRNA is clean- to avoid toxic effects. so many folks choose HPLC to get rid of salts etc
