I have to order dsRNA (25nt) and I asked for a quote. The supplier insert HPLC purification that is very expensive. I would like to know if it's strictly necessary. No HPLC purification would affect negatively my experiment?
dsRNAs with 25nt need to be purified. The batch synthesis (unpurified) may not have better yield hence may impair with your experimental outcome.
if you just want to produce siRNA- no need for HPLC purification, just mix strands in 1:1 ratio and thats it. small excess of one strand wont make a difference. for in vivo studies of course you want to ensure your siRNA is clean- to avoid toxic effects. so many folks choose HPLC to get rid of salts etc
no need of purification ...I simply use desalted siRNA . it work great .
in europe: eurogentec, eurofins provide good quality siRNA
beware of what the company claim..if they have real synthesis skills

yes, desalting is enough ...I never use HPLC purified siRNA...better save money for other reagents..:-)
