I am now working with the HUVEC cell line. I made my complete media this way like F12K nutrient mix with 10% FBS, 0.1 mg/mL heparin sodium salt, and 0.03 mg/mL endothelial cell growth supplement (ECGS).To avoid contamination, I used 50 mg/mL of Geneticin instead of Gentamycin at the same concentration. However, after seeding the cells into the culture flask, I discovered that they were all dead on next day. That's why I rethawed a new vial from stock, this time I use Penstrep as the antibiotic for making complete media, and my HUVEC cells are now attached to the surface and growing well.
Please offer some ideas.
