I'm using stable transfected CHO DG44 cells with geneticin and dhfr (double transfection) as selection markers. Since MTX is used for gene amplification of the dhfr gene, is it useful to increase G418 concentration to amplify the neo resistence gene in order to amplify the other expression cassette? (I want to have a similar expression rate for both POI...)
You could do in a similar way as MTX. You should star with the concentration that kill your cells in 15 days and then progressively increase the concentration. Before you increase the concentration of your cells keep cells from previous step. You cloud also select the levels of expression that is optimal for your work by cloning (if you have a detection system as ELISA). Check that the growth of you cells is not affected.
